Position:Senior Research AssociateDuties:Research the effects of substances on biological processes by running routine cell-based structure-activity relationship assays by typically using flow cytometry assays on human whole blood or cell samples treated with Arcus compounds. Analyze and prepare reports based on research data, including curve-fitting and IC50 determination. Work with other Scientists and Research Associates to develop and perform additional assays using various formats such as PCR, ELISA and luminescence detection as needed.

Provide live cell stocks of up to 25-35 mammalian cell lines daily for various projects and assays run in Biology, Discovery Pharmacology, and DMPK groups. Provide bulk cell line preps for use in animal model studies as well as scale up and freeze aliquots of new cell lines for banking. Isolate and expand single cell clones of stably transfected cells and test clones for function using FACS (fluorescence-activated cell sorting) or other functional assays in collaboration with the Cell Engineering group. Perform PCR (polymerase chain reaction) and luminescence-based mycoplasma testing on cell lines being grown by various researchers.Implement a new electronic notebook-based inventory for the Central Cell Line Banking function and train researchers throughout the company in its useAssist researchers with selecting appropriate cell lines based on species, tissue, and disease and then deliver frozen cell stocks to researchers and update records for the central cell bank.

Stock media, reagents, and consumables for all cell culture labs in various departments in collaboration with the Lab Manager as well as test lots of human and fetal bovine serum for cell growth prior to bulk purchases. Assist with maintenance of lab equipment, including automated cell counters, incubators, milliQ water systems, robotics and liquid handlers, plate readers, and flow cytometers.EducationRequired:Master or foreign equivalent in Pharmacology, Cell Biology, Biochemistry or related field.ExperienceRequired:3 years of experience as Research Assistant, Research Analyst or related occupation.Special Requirements:Must have at least 1 year of prior work experience in each of the following:

  • Culturing various cancer cell lines. Provide live cell stocks of up to 25-35 mammalian and animal cell lines daily for various projects, assays and animal studies run in Biology, Discovery Pharmacology, and DMPK groups. Scale up and freeze aliquots of new cell lines for banking.
  • Run routine cell-based structure-activity-relationship assays using flow cytometry on human whole blood samples and other cells treated with compounds.
  • Antibody staining techniques, and use of FACS CANTO, Attune flow cytometry and Automated liquid handling instruments.
  • Analysis and reporting of data, including curve fitting and IC-50 determination, using data analysis software tools including FlowJo, GraphPad Prism and Microsoft suite.
  • Databases to implement a new electronic notebook-based inventory for the Central Cell Line Banking function.
  • Biochemical and cell-based assay development for compound screening using Invitro assays including Elisa, PCR and other immunoassay methods.
  • Manage daily activities of a cell core laboratory including purchasing and maintenance of the lab equipment.
